油桃组蛋白基因PpH3.3的克隆和表达分析

摘 要:

组蛋白H3与H2A、H2B、H4共同形成真核生物染色质核小体的八聚体核心。本文通过RACE的方法从休眠期的油桃花芽中克隆到一个编码组蛋白H3变体的基因, 命名为PpH3.3。序列分析结果表明, 该基因全长821 bp, 存在一个完整的开放阅读框411 bp, 编码136个氨基酸。预测的氨基酸序列具有H3家族典型的保守结构域, 与拟南芥、葡萄的组蛋白H3, 具有较高的相似性。利用荧光定量PCR技术分析该基因在花芽休眠进程中的表达特异性, 发现PpH3.3在休眠期表达水平显著高于休眠诱导期和休眠解除期, 并且单氰胺和高温处理能够诱导类似的表达模式。

Clone and Expression Analysis of a cDNA Encoding Histone H3.3 in Prunus persica var. nectarina Maxim

Abstract:

Histone H3 is one of the four histones, along with H2A, H2B and H4, which forms the eukaryotic nucleosome octomer core. In this study, a novel histone H3 variant gene was isolated from dormant Prunus persica var. nectarina floral buds with rapid amplification of cDNA ends (RACE), designated as PpH3.3. The PpH3.3 cDNA is 821 bp long and contains an open reading frame (ORF) of 411 bp encoding 136 amino acids. Sequence alignments indicate that PpH3.3 contains conserved regions found in histone H3 and shows a high level of sequence similarity to many other plants. The expression profile of the PpH3.3 gene detected by QRT-PCR showed that the transcripts of PpH3.3 was high in dormancy period of buds and both high temperature and hydrogen cyanamide lead similar expression pattern.
